Тем не менее стволовые клетки миокарда детей являются практически неизученными. Целью работы было получение стволовых клеток миокарда из небольших (1-3 мм) образцов тканей от пациентов детского возраста, характеристика этих клеток, сравнение их со стволовыми клетками миокарда взрослого в отношении их пролиферативного и дифференцировочного потенциала. Из образцов детского миокарда, полученных в ходе коррекции врожденных пороков сердца, путем ферментативной диссоциации ткани получали линии пролиферирующих клеток; проводили их фенотипирование при помощи проточной цитометрии, методов полуколичественной и количественной полимеразной цепной реакции; оценку их пролиферативного и дифференцировочного потенциала при помощи индукции дифференцировки в кардиогенном, направлениях. Было получено пять линий стволовых клеток сердца. Полученные линии интенсивно пролиферировали, экспрессировали маркеры кардиогенного происхождения и имели потенциал к дифференцировке в нескольких направлениях. Полученные клетки были сходны по характеристикам с описанными ранее клетками, получаемыми из миокарда взрослого человека. Результаты проведенного исследования указывают на то, что существует возможность получения линий клеток из небольших образцов ткани детского миокарда, остающихся в ходе операций по коррекции врожденных пороков сердца. Эти клетки представляют собой гетерогенную популяцию, имеющую мультипотентные свойства, имеют фенотипические характеристики, сходные с клетками, получаемыми из миокарда взрослого, высокий регенеративный потенциал, способность к кардиогенной дифференцировке.</p></abstract><trans-abstract xml:lang="en"><p>Myocardial progenitor cells represent a perspective source for cell therapy of cardiovascular disorders and are intensively studied. However, mainly the cells from adult patients are studied whereas the cells derived from children with congenital heart diseases remained poorly investigated. The aim of the present work was to obtain progenitor cells from intraoperatively obtained cardiac tissue from infants and children undergoing repair of congenital cardiac defects. Following isolation, the cells gave rise to a clonogenic, highly proliferative spindle-shaped cell population. The cells expressed markers of cardiogenic origin and were shown to differentiate towards cardiogenic lineage. This resident myocardial progenitor cells obtained from infant myocardial tissue demonstrate similar characteristics to previously described cells derived from adult myocardial tissue.
